Vesicle Tracing

1. Open up Vast, the EM you want, and a new segmentation.
2. Set auto fill(the check box labeled ‘Fill’) and conditional painting. Both of these are located in the options to the right.
3. A good range for conditional painting is 0.00 - 0.34.
4. Begin tracing by looking for vesicle clouds and find the synapse.
5. If a cloud has no synapse there is no need to trace it.
6. Trace the whole cloud as long as it’s continuous.
7. Be careful of boundaries and other organelles that might get caught in the conditional tracing.

Vesicle Reviewing

1. Open the corresponding synapse segmentation for your EM volume.
2. Check how many synapses have been traced with how many vesicles clouds have traced.
3. If either segmentation (vesicle or synapse) has fewer segments try to locate where we’ve missed one. (Cloud or clef.)
